● 可以將逆轉(zhuǎn)錄病毒中的目的基因高效轉(zhuǎn)入造血干細(xì)胞、T淋巴細(xì)胞等哺乳動物來源的細(xì)胞。 ● 避免感染過程中因使用polybrene造成的細(xì)胞毒作用。 ● 同樣適用于感染常規(guī)的細(xì)胞系。 ● RetroNectin是重組人纖維連接蛋白片段,包括細(xì)胞結(jié)合域,肝磷脂結(jié)合域和CS1位點三個功能區(qū)域。它由574個氨基酸組成,分子量63 kDa。RetroNectin能夠大大的提高逆轉(zhuǎn)錄病毒感染哺乳動物細(xì)胞的感染效率,其作用原理是:細(xì)胞表面VLA-4及VLA-5可分別于RetroNectin上的CS-1位點及細(xì)胞結(jié)合域結(jié)合,逆轉(zhuǎn)錄病毒載體可以與肝磷脂結(jié)合域結(jié)合。在經(jīng)RetroNectin包被的培養(yǎng)容器表面,細(xì)胞與病毒載體以高濃度共存,從而提高基因轉(zhuǎn)染效率30%-70%。 ● 另外RetroNectin還可以刺激淋巴細(xì)胞增殖,尤其是胸腹水以及腫瘤組織分離的TIL細(xì)胞,利用RetroNectin技術(shù)均可使其獲得上千倍的增殖,已有部分客戶將RetroNectin技術(shù)擴(kuò)增TIL的方法進(jìn)行專利申請。 |
