Cellartis 2i mES/iPSC Culture Medium是一款無血清的、成分確定的細胞培養(yǎng)基,可用于小鼠ES和iPS細胞建系。該培養(yǎng)基含有兩種小分子抑制劑(基于2i技術(shù):CHIR99021、PD0325901),分別是用于抑制GSK3β和ERK/MEK介導的分化誘導信號通路,以及促進細胞增殖。
單獨使用Cellartis 2i mES/iPSC Culture Medium,可以長期維持小鼠ES細胞處于原始狀態(tài)和基態(tài)(Naive, ground state)或者小鼠iPS細胞處于完全多能性(Fully pluripotent)的狀態(tài)。添加LIF(Leukemia Inhibitory Factor),可以通過調(diào)節(jié)Nanog信號通路使得Partial-iPS細胞或Pre-iPS細胞有效轉(zhuǎn)化為具有完全多能性的iPS細胞(Silva et al. 2008; Silva et al. 2009).
2i技術(shù)也應用于從難獲得具有生殖系能力(germline-competent)ES細胞的小鼠品系中建立ES細胞系(Nichols et al. 2009),以及用于從人類和家畜物種中誘導iPS細胞(Wang et al. 2011; Malaver-Ortega et al. 2012)。 |
